AbstractA new approach of preparing monolithic silica column is reported, in which an improved sol-gel technology was adopted with tetramethoxysilane and polyethylene glycol as the matrix materials. The problems of crack and deformation in preparing the column were solved by using N,N-dimethylformamide and a heat treatment procedure. The methods of mesopore fabrication and column back pressure as a function of the flow-rate on the monolithic silica column were investigated, and the monolithic silica column was applied to separation of benzene substituted compounds which were baseline-separated in normal-phase chromatography. The results showed that the column prepared possessed bimodal pores structures with micrometer-size co-continuous through-pores and nanometer-size mesopores, straight appearance, high permeability and low column back pressure, high mechanical strength and column efficiency, which are well suited for high speed and high efficiency separations and analyses for compounds.
