Studies on preparation and properties of polyprrole uricase electrode

AbstractThree types of polypyrrole uricase electrodes were prepared by means of the spontaneous adsorption of uricase at open circuit, immobilization of uricase at an applied negative potential or at an applied positive potential respectively on the polypyrrole films. The initial response current of the enzyme electrode formed at the positive potential was the highest and that of the enzyme electrode formed at the negative potential was the loa est among the three types of electrodes. The response current of the enzyme electrode formed at positive potential increased with time first, thee decreased and slowly to form a current platform. The effective life. time of this electrode was over 60 days. The XPS experiments reveal that a peak of C-ls with 285.9 eV for the polypyrrole film, and two peaks of C-ls with 285.9 eV and 288.9 eV for the freshly prepared polypyrrole uricase electrode were observed. The latter was caused by the chemical shift in XPS of C-ls in polypyrrole due to the effect of the immobilized uricase, The pH effect on the maximum reaction rate (i.e. maximum response current) of the enzyme electrode indicates that the immobilized uricase has a basic ionizing group involved in the catalytic activity.
